Biological Activity

RP-64477 is a potent inhibitor of acyl-coenzyme A:cholesterol O-acyltransferase (ACAT), an enzyme central to intracellular cholesterol esterification. By inhibiting ACAT activity across intestinal, hepatic, and monocytic cell models, RP-64477 suppresses the esterification of free cholesterol into cholesteryl esters. This inhibition impairs cellular lipid storage, reduces apolipoprotein B secretion, and prevents foam cell formation, thereby altering cellular cholesterol homeostasis and lipid transport pathways. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
