Rogaratinib (BAY-1163877) is a potent pan-FGFR inhibitor that targets FGFR1 (IC50 1.8 nM), FGFR2 (IC50 < 1 nM), FGFR3 (IC50 9.2 nM), and FGFR4 (IC50 1.2 nM). By blocking receptor autophosphorylation, it suppresses downstream FGFR signaling pathways, including MAPK and AKT/mTOR. This inhibition leads to reduced cell proliferation and induces apoptosis in cancer models harboring FGFR genetic alterations or elevated mRNA expression. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
