Ro 20-1724 is a selective inhibitor of phosphodiesterase 4 (PDE4) with a Ki of 3.1 µM for human PDE4. By preventing intracellular cyclic AMP (cAMP) degradation, Ro 20-1724 elevates cAMP levels and enhances cAMP-dependent signaling. This pathway activation regulates downstream physiological functions and stimulates histamine-mediated gastric acid secretion, as demonstrated by increased aminopyrine accumulation in gastric glands. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
