RMC-4550 is a potent and selective allosteric inhibitor of SHP2 (IC50 = that displays minimal activity against a panel of 468 kinases and 15 phosphatases. By stabilizing the inactive conformation of SHP2, RMC-4550 inhibits SHP2-mediated RAS nucleotide exchange, leading to reduced RAS-GTP levels and suppression of downstream MAPK/ERK pathway signaling to impair cancer cell growth. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
