Biological Activity

RK-701 is a potent, selective inhibitor of histone methyltransferases G9a and GLP1. It suppresses histone methyltransferase activity and inhibits the recruitment of γ-globin repressors in complex with G9a through CHD4, a component of the NuRD complex. This interaction selectively upregulates BGLT3 long non-coding RNA and induces fetal γ-globin expression in human erythroid cells and mouse models, supporting research in sickle cell disease. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
