RJG-2051 is a selective covalent sulfur-triazole exchange (SuTEx) inhibitor targeting aldo-keto reductase family 1 member C3 (AKR1C3) at a noncatalytic tyrosine residue (Y24). By covalently modifying the Y24 site, RJG-2051 effectively blocks the enzymatic activity of AKR1C3, thereby disrupting key downstream signaling and cellular metabolic pathways involved in androgen, estrogen, and prostaglandin biosynthesis.
