Biological Activity

Ripretinib (DCC-2618) is a switch-pocket control inhibitor targeting wild-type KIT and KIT mutants, including KIT V654A, KIT T670I (IC50 1, KIT D816H, and KIT D816V (IC50 1, as well as PDGFRα. By locking the kinase in an inactive conformation, it blocks downstream KIT and PDGFRα signaling pathways, inhibiting cell proliferation and survival in driven tumor cell models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
