Biological Activity

Rilpivirine Hydrochloride (TMC278) is a second-generation non-nucleoside reverse transcriptase inhibitor (NNRTI) targeting HIV-1 reverse transcriptase. By binding to the non-nucleoside inhibitor-binding pocket, it inhibits viral RNA-dependent DNA polymerase activity, thereby blocking viral replication and viral DNA synthesis in wild-type and NNRTI-resistant HIV-1 strains. Additionally, rilpivirine acts as an inhibitor of ATP-binding cassette transporters, including MDR1 and BCRP. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
