Biological Activity

Rezatapopt (PC14586) is a first-in-class, selective small-molecule reactivator of the Y220C-mutated p53 protein. By binding to a unique mutation-created pocket, it stabilizes the wild-type conformation of p53 Y220C, restoring its DNA-binding ability and transcriptional activity. Reactivation of the p53 signaling pathway restores downstream tumor suppressor functions, driving transcriptomic target activation, antiproliferative responses, and significant tumor growth inhibition or regression in p53 Y220C-mutant xenograft models.[1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
