Biological Activity

RA-190 is an orally active bis-benzylidene piperidone that serves as a potent and selective inhibitor of the proteasome ubiquitin receptor RPN13 (ADRM1). By binding to RPN13, RA-190 impairs proteasomal degradation of toxic polyubiquitinated proteins, which triggers an endoplasmic reticulum (ER) stress response, activates the downstream p53/p21 signaling axis, and ultimately promotes autophagy and apoptosis in multiple myeloma cells.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
