(R)-Nepicastat HCl is a potent and selective dopamine-β-hydroxylase inhibitor, demonstrating inhibitory activity against bovine dopamine-β-hydroxylase (IC50 25.1 nM) and human dopamine-β-hydroxylase (IC50 18.3 nM). By inhibiting dopamine-β-hydroxylase, the enzyme responsible for converting dopamine to norepinephrine, the compound disrupts catecholamine biosynthesis. This inhibition decreases tissue norepinephrine levels while elevating dopamine concentrations, thereby attenuating sympathetic neurotransmission and modulating downstream cardiovascular and neurochemical signaling pathways.
