(R)-3-Hydroxybutanoic acid (D-3-hydroxybutyrate) is an endogenous ketone body and an endogenous inhibitor of histone deacetylases (HDACs), including HDAC1 and HDAC3. It acts through epigenetic modulation to enhance histone acetylation and upregulate antioxidant gene expression, suppressing oxidative stress in mammalian cells. Additionally, (R)-3-hydroxybutanoic acid serves as a signaling metabolite and selective agonist for hydroxycarboxylic acid receptor 2 (HCAR2/GPR109A), regulating metabolic homeostasis and lipolysis. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
