Biological Activity

PYZD-4409 is a small-molecule inhibitor of the ubiquitin-activating enzyme UBA1 (E1) with an IC50 of. By blocking UBA1, PYZD-4409 impairs ubiquitin conjugation, triggering endoplasmic reticulum stress and accumulation of misfolded proteins. This leads to the induction of cell death in malignant cells and preferentially inhibits the clonogenic growth of primary acute myeloid leukemia cells, presenting a potential therapeutic strategy for hematologic malignancies. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
