Biological Activity

Pyrvinium pamoate is an anthelmintic agent and potent allosteric activator of casein kinase 1α (CK1α) that functions as a Wnt/β-catenin signaling pathway inhibitor. By selectively activating CK1α, pyrvinium pamoate accelerates the phosphorylation and degradation of β-catenin, thereby repressing Wnt target gene transcription. This downregulates downstream oncogenic drivers and suppresses cell proliferation, migration, and survival in various Wnt-dependent cancer cell lines and tumor models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
