Pyranine (HPTS) is a cell-impermeant, pH-sensitive fluorescent indicator and hydrophilic optical detecting reagent. It functions as a chemosensor for pH and copper ions (Cu⁺), enabling fluorescence-based optical detection of proton gradients and endosomal acidification. Upon excitation, its pH-dependent fluorescence spectra allow non-invasive quantitative monitoring of intracellular pH changes and endocytic compartment dynamics in biological systems without traversing intact plasma membranes.
