PYR-41 is a cell-permeable inhibitor of ubiquitin-activating enzyme E1 (UBA1) that shows no inhibitory activity toward ubiquitin-conjugating enzymes (E2). By blocking ubiquitin E1 function, PYR-41 prevents ubiquitination and ubiquitin-mediated protein degradation, leading to attenuated NF-kB activation and stabilization of tumor suppressor proteins such as p53. Consequently, PYR-41 disrupts cellular protein homeostasis and effectively induces apoptosis in cancer cells. [1]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
