PX-12 is an irreversible thioredoxin-1 (Trx-1) inhibitor that acts through thioalkylation of the Cys73 residue. By blocking Trx-1 redox activity, PX-12 disrupts cellular redox balance, leading to decreased levels of hypoxia-inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF). Consequently, PX-12 inhibits tumor cell proliferation, impairs angiogenesis, and suppresses tumor growth in human cancer xenograft models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
