Biological Activity

Purvalanol B is a potent, selective cyclin-dependent kinase inhibitor targeting cdc2-cyclin B (IC50 = 6 nM), CDK2-cyclin A (IC50 = 6 nM), CDK2-cyclin E (IC50 = 9 nM), and CDK5-p35 (IC50 = 6 nM). By inhibiting cyclin-dependent kinase complexes, it disrupts cell cycle progression at the G1/S and G2/M transitions, leading to cell cycle arrest and induction of apoptosis in mammalian cells. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
