PTC-028 is an orally bioavailable small-molecule inhibitor of BMI-1 that decreases BMI-1 protein levels through posttranslational hyperphosphorylation and subsequent proteasomal degradation. By targeting BMI-1, PTC-028 impairs the function of polycomb repressive complex 1 (PRC1), thereby inhibiting cancer stem cell self-renewal, suppressing tumor cell proliferation, and triggering cell cycle arrest and apoptosis in preclinical epithelial ovarian cancer models.
