Biological Activity

PT2399 is a potent and selective HIF-2α antagonist that directly binds to the HIF-2α PAS-B domain. By disrupting the heterodimerization of HIF-2α with HIF-1β, PT2399 inhibits HIF-2-mediated transcriptional activity and downregulates downstream target genes involved in hypoxia signaling. Consequently, this inhibition suppresses tumor cell proliferation and demonstrates robust preclinical antitumor efficacy in clear cell renal cell carcinoma models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
