Biological Activity

PS47 is an inactive E-isomer of PS48, functioning as a negative control compound for the allosteric activation of 3-phosphoinositide-dependent protein kinase 1 (PDPK1). Unlike PS48, PS47 fails to bind the hydrophobic motif PIF-pocket or induce allosteric conformational changes at the ATP-binding site of PDPK1. Consequently, it does not activate downstream PI3K/AKT pathway signaling, serving as a specificity tool in kinase studies.[1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
