Biological Activity

PS210 is a substrate-selective, allosteric activator of 3-phosphoinositide-dependent protein kinase 1 (PDK1) that targets the hydrophobic PIF-pocket docking site. Binding of PS210 to the PIF-pocket allosterically modulates PDK1 active site conformation, enhancing kinase activity toward substrates lacking a PIF motif while inhibiting phosphorylation of distinct targets. This selective activity modulates downstream PI3K/AKT signaling pathways to influence cellular survival and metabolic responses. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
