Biological Activity

Prucalopride (R-93877) is a selective, high-affinity 5-HT4 receptor agonist that displays high selectivity over other 5-HT receptor subtypes. Activation of 5-HT4 receptors stimulates adenylate cyclase and elevates intracellular cyclic AMP levels, triggering acetylcholine release in enteric neurons. This cholinergic pathway stimulation promotes gastrointestinal motility, enhances peristaltic reflexes, and facilitates intestinal transit without significant cross-reactivity at other monoamine receptors.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
