PRT543 is a potent small-molecule inhibitor of protein arginine methyltransferase 5 (PRMT5). Pharmacologic inhibition of PRMT5 downregulates DNA damage repair (DDR) and DNA replication pathway genes—including BRCA1/2, RAD51, and ATM—by altering chromatin methylation and alternative splicing. Consequently, PRT543 suppresses cell proliferation, demonstrates antitumor activity in adenoid cystic carcinoma, and inhibits the growth of PARP inhibitor-resistant tumors in vivo. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
