Biological Activity
PRT3645 is a brain-penetrant small-molecule inhibitor targeting cyclin-dependent kinases CDK4 and CDK6 with high selectivity over other CDK family members. By directly inhibiting CDK4/6 complexes, PRT3645 blocks retinoblastoma protein phosphorylation and arrests cell cycle progression from G1 to S phase. This inhibition disrupts downstream oncogenic pathway signaling, leading to suppressed cell proliferation and robust anti-tumor activity in glioblastoma and intracranial breast cancer metastasis models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
