Biological Activity

PROTAC SGK3 degrader-1 is a proteolysis-targeting chimera (PROTAC) designed to target serum/glucocorticoid-regulated kinase 3 (SGK3) for von Hippel-Lindau (VHL)-mediated proteasomal degradation. By conjugating an SGK inhibitor to a VHL ligand, it selectively recruits SGK3 to the E3 ubiquitin ligase complex for degradation. This depletion suppresses downstream PI3K/Akt and SGK signaling pathways, leading to inhibited cell proliferation in SGK3-dependent cancer cell lines.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
