PRN1371 is a potent, irreversible covalent inhibitor targeting fibroblast growth factor receptors FGFR1, FGFR2, FGFR3, and FGFR4, along with CSF1R. By covalently binding to the active kinase domain, PRN1371 effectively disrupts receptor autophosphorylation and downstream MAPK and AKT/mTOR signaling pathways. Consequently, this target inhibition suppresses cellular proliferation, blocks cell cycle progression, and reduces tumor growth in preclinical FGFR-dependent disease models. [1]
