Prifemilast is a potent and selective phosphodiesterase 4 (PDE4) inhibitor. By inhibiting PDE4 enzymes, it prevents the breakdown of cyclic adenosine monophosphate (cAMP), leading to elevated intracellular cAMP levels. This elevation suppresses inflammatory signaling pathways and downregulates the production of pro-inflammatory cytokines, resulting in significant anti-inflammatory and anti-pruritic effects in preclinical models of skin inflammation.
