Biological Activity

Preladenant is a potent, competitive, and selective antagonist of the human adenosine A2A receptor with a Ki of 1.1 nM. By blocking adenosine A2A receptor activation, Preladenant inhibits cAMP accumulation and downstream PKA signaling, modulating GABAergic and dopaminergic neurotransmission in the striatum. In preclinical models, Preladenant demonstrates potent neuroprotective and antiparkinsonian effects, improving motor deficits without inducing dyskinesia. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
