Biological Activity

Pradigastat is a potent and selective inhibitor of human diacylglycerol acyltransferase 1 (DGAT1) that also inhibits uptake transporters OATP1B1, OATP1B3, and OAT3. By inhibiting DGAT1, pradigastat blocks the final step of triacylglycerol biosynthesis, which impairs intestinal chylomicron assembly and suppresses postprandial hypertriglyceridemia. Additionally, its action on OATP and OAT uptake transporters alters solute transport pathways involved in hepatic and renal clearance. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
