Biological Activity

PQ 401 is a diarylurea small-molecule inhibitor of insulin-like growth factor-1 receptor (IGF-1R) that inhibits IGF-1R domain autophosphorylation with an IC50 of <1 μM. By blocking IGF-1R kinase activation, it impairs downstream AKT/mTOR and MAPK signaling pathways. This pathway inhibition suppresses cellular proliferation, induces apoptosis, and attenuates overall tumor cell survival. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
