Biological Activity

Poloxin is a non-ATP competitive inhibitor of Polo-like kinase 1 polo-box domain (Plk1 PBD) with an apparent IC50 of. It displays selectivity over Plk2 PBD (IC50 approximately 4-fold higher) and Plk3 PBD (IC50 approximately 11-fold higher). By disrupting Plk1 PBD-mediated substrate binding, Poloxin impairs mitotic spindle assembly, ultimately inducing mitotic arrest and apoptosis in cancer cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
