PNU-120596 is a selective positive allosteric modulator of α7 neuronal nicotinic acetylcholine receptors (α7 nAChR) with an EC50 of 216 nM. It exhibits no detectable activity against α4β2, α3β4, and α9α10 nicotinic receptors. Functionally, PNU-120596 enhances agonist-evoked calcium flux and increases peak currents while significantly prolonging agonist-induced channel responses, thereby potently enhancing α7 nAChR-mediated cholinergic signaling.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
