Biological Activity

Plixorafenib (PLX8394) is an orally available, small-molecule kinase inhibitor targeting BRAF(V600E), wild-type BRAF, and CRAF. It selectively blocks both monomeric and dimeric BRAF signaling without triggering paradoxical MAPK/ERK pathway activation in RAS-mutant cells. By potently inhibiting downstream MEK and ERK phosphorylation, PLX8394 disrupts oncogenic signaling cascades, leading to effective suppression of cancer cell proliferation and inhibition of tumor growth in BRAF-driven malignancies.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
