Biological Activity

AMD3100 (Plerixafor) is a potent chemokine receptor antagonist that selectively targets CXCR4 to inhibit CXCL12-mediated chemotaxis. By blocking the binding of CXCL12 to CXCR4, AMD3100 disrupts downstream CXCL12/CXCR4 signaling pathways, impairing intracellular calcium mobilization and cell migration. Consequently, this compound suppresses CXCR4-dependent human immunodeficiency virus (HIV) replication and promotes the mobilization of hematopoietic stem and progenitor cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
