Biological Activity

Pivmecillinam hydrochloride is an orally bioavailable prodrug of mecillinam, a β-lactam antibiotic with selective activity against Gram-negative bacteria. Upon conversion to mecillinam, it specifically targets and inhibits penicillin-binding protein 2 (PBP-2), disrupting bacterial cell wall biosynthesis and peptidoglycan cross-linking. This inhibition impairs cell elongation, inducing spherical cell formation and ultimately leading to bacterial cell lysis. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
