Pitolisant (BF2.649) is a potent, selective antagonist and inverse agonist of the histamine H3 receptor, exhibiting a Ki value of 0.16 nM and an EC50 of 1.5 nM. By blocking presynaptic H3 autoreceptors, it increases histamine release in the central nervous system, thereby enhancing histaminergic neuronal signaling and promoting vigilance and cognitive function in research models of wakefulness and memory deficits.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
