Pinometostat (EPZ5676) is an S-adenosyl methionine-competitive inhibitor of protein methyltransferase DOT1L (Ki 80 pM) with over 37,000-fold selectivity against other histone methyltransferases. By blocking DOT1L activity, it inhibits histone H3 lysine 79 (H3K79) methylation and suppresses leukemogenic gene expression, resulting in selective inhibition of cell proliferation and induction of apoptosis in MLL-rearranged leukemia cells. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
