Biological Activity

Pifithrin-β (cyclic pifithrin-α) is a small-molecule inhibitor of the tumor suppressor p53 transcription factor [1, 2]. By suppressing p53-dependent transcriptional activation, it downregulates downstream targets like p21 and counteracts p53-mediated cell cycle arrest and apoptosis [1, 2]. In wild-type p53 tumor cells, cyclic pifithrin-α sensitizes cells to antimicrotubule agent-induced apoptotic cell death [1] and mitigates cell death driven by toxic stressors in neurodegenerative models [2].[1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
