Pico145 is a selective TRPC1/4/5 channel inhibitor that inhibits (-)-englerin A-activated TRPC4 (IC50 0.349 nM) and TRPC5 (IC50 1.3 nM). By potently blocking TRPC1/4/5 channel complexes, it impairs store- and receptor-operated calcium influx pathways, suppressing downstream calcium signaling. Furthermore, Pico145 exhibits high selectivity over other ion channels, showing no inhibitory effect on TRPC3, TRPC6, TRPV1, TRPV4, TRPA1, TRPM2, or TRPM8 channels.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
