PI-273 is a substrate-competitive, subtype-specific phosphatidylinositol 4-kinase (PI4KIIα) inhibitor with an IC50 of 0.47 μM. By directly binding PI4KIIα, PI-273 reduces cellular phosphatidylinositol 4-phosphate (PI4P) levels and attenuates AKT signaling pathway activity. Downstream, this selective inhibition retards cell proliferation by inducing G2/M phase cell cycle arrest, suppressing colony formation, and promoting cell apoptosis in breast cancer cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
