PhIP (2-Amino-1-methyl-6-phenylimidazo[4,5-b]pyridine) is a carcinogenic heterocyclic aromatic amine and estrogen receptor alpha (ERα) agonist. Following metabolic activation by CYP1A2 into reactive hydroxylated intermediates, PhIP induces C8-dG adduct formation and genomic DNA damage. Independently of genotoxicity, PhIP binds ERα to activate estrogenic signaling, promoting transcriptional changes and cell proliferation in breast cancer cell models. [1]
