Biological Activity

PHD-1-IN-1 is a potent and orally active inhibitor targeting hypoxia-inducible factor prolyl hydroxylase domain-1 (PHD-1) [1]. It binds to the active site Fe2+ ion through a monodentate interaction, suppressing PHD-1-mediated proline hydroxylation [1]. Inhibition of PHD-1 prevents oxygen-dependent degradation of hypoxia-inducible factors, thereby regulating downstream HIF signaling pathways involved in cellular hypoxic responses and gene transcription. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
