Milciclib (PHA-848125) is an ATP-competitive cyclin-dependent kinase inhibitor targeting cyclin A/CDK2 and tropomyosin receptor kinase A (TRKA). It suppresses retinoblastoma protein phosphorylation, reduces cyclin A expression, and upregulates p21, p27, and p53 expression. These actions trigger G1 phase cell cycle arrest and induce cell death through autophagy. Additionally, Milciclib displays significant antiproliferative activity and antitumor efficacy across various preclinical carcinoma xenograft models. [1]
