Biological Activity

PFI-2 HCl is a potent, selective, substrate-competitive lysine methyltransferase SETD7 inhibitor with an IC50 of 2 nM and an apparent Ki of 0.33 nM. It exhibits greater than 1000-fold selectivity over other methyltransferases and non-epigenetic targets. Inhibition of SETD7 by PFI-2 alters the Hippo pathway signaling network by suppressing Hippo pathway co-activator YAP transactivation and modulating downstream gene expression profiles. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
