PF 429242 is a selective site-1 protease (S1P) inhibitor (IC50 = 170 nM) that displays high selectivity over other serine proteases. By inhibiting S1P, PF 429242 blocks the endoproteolytic cleavage and activation of sterol regulatory element-binding proteins (SREBPs). This disruption of the SREBP signaling pathway suppresses downstream lipogenic target gene expression, ultimately leading to reduced cellular cholesterol and fatty acid synthesis.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
