Biological Activity

PF-6260933 is a potent inhibitor of MAP4K4 with high kinome selectivity. By selectively targeting MAP4K4 kinase activity, this compound suppresses downstream signaling cascades through the JNK pathway. Consequently, MAP4K4 inhibition regulates key cellular and vascular phenotypes, inhibiting cell migration, protecting endothelial function, and attenuating pro-inflammatory gene expression in cell-based models.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
