Biological Activity

PF-06952229 is a selective, orally bioavailable small-molecule inhibitor of TGF-β receptor 1 (TGF-β-R1/ALK5). By blocking TGF-β-R1 kinase activity, it suppresses SMAD2 phosphorylation in both tumor and immune cells, thereby interrupting canonical TGF-β signaling and reversing epithelial-mesenchymal transition in vitro. Consequently, PF-06952229 modulates the tumor immune microenvironment and demonstrates robust antitumor activity in preclinical mouse models.[1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
