PF-06726304 is a potent and selective inhibitor of enhancer of zeste homolog 2 (EZH2), targeting both wild-type EZH2 and the Y641N mutant isoform. By inhibiting EZH2 methyltransferase activity, it suppresses trimethylation of histone H3 at lysine 27 (H3K27me3). Decreased H3K27me3 levels lead to derepression of silenced tumor suppressor genes, promoting cell cycle arrest, transcriptional reprogramming, and suppression of tumor cell proliferation in EZH2-dependent malignancies.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
