PF-06424439 Methanesulfonate is an orally bioavailable inhibitor of diacylglycerol acyltransferase 2 (DGAT2) with an IC50 of 14 nM. By inhibiting DGAT2, it disrupts the final step of triacylglycerol synthesis, blocking the esterification of diacylglycerol with fatty acyl-CoA. This targeted inhibition reduces intracellular triglyceride accumulation, decreases hepatic lipid droplet formation, and suppresses triglyceride secretion in metabolic disease models. [1]
